Biosynthesis of Gamma-Aminobutyric Acid by Engineered Clostridium tyrobutyricum Co-Overexpressing Glutamate Decarboxylase and Class I Heat Shock Protein

Gamma-aminobutyric acid (GABA) is a major inhibitory neurotransmitter in the mammalian central nervous system that has significant beneficial effect on human health. Traditional microbial GABA synthesis requires continuous oxygen supplementation. Here, new anaerobic platform for production was established with engineered C. tyrobutyricum ATCC 25755, which considered an ideal microbial-cell factory bioproduction. Glutamate decarboxylase (GAD) and Class I heat-shock proteins were screened overexpressed, generating excellent Ct-pMAG strain monosodium-glutamate (MSG) tolerance production, titer of 14.26 g/L serum bottles mixed substrate glucose MSG. Fed-batch fermentation carried out 5 L bioreactor, achieving 35.57 122.34 final titers by applying pH-free strategy pH-control strategy, respectively using Finally, two-stage (growth stage bioconversion stage) applied glutamate (L-Glu) as substrate, obtaining 400.32 productivity 36.39 g/L/h. Overall, this study provides anaerobic-fermentation high-level bio-GABA production.
